چکیده

SOD2 encodes manganese superoxide dismutase, a mitochondrial enzyme that protects the cell against damage from superoxide free radicals. A common polymorphism, a T ! C transition (Val16Ala), was shown to alter the manganese superoxide dismutase mitochondrial targeting sequence (1). Animal studies have linked reduction in manganese superoxide dismutase activity to increased DNA damage and higher incidence of cancer (2). NQO1 encodes NAD(P)H dehydrogenase (quinone), a flavoprotein that also protects the cell against cytotoxicity, by promoting the two-electron reduction of quinoid compounds to hydroquinones (3). A common polymorphism, a C ! T transition (Pro187Ser), produces significant reductions in enzyme activity in human colon cancer cells (3). Therefore, in the case-control study reported here, we examined the associations between polymorphisms in SOD2 and NQO1 , cigarette smoking, and bladder cancer risk.

Genetic polymorphisms in NQO1 and SOD2: interactions with smoking, schistosoma infection, and bladder cancer risk in Egypt.

BACKGROUND Bladder cancer is the most prevalent form of cancer in men among Egyptians, for whom tobacco smoke exposure and Schistosoma haematobium (SH) infection are the major risk factors. We hypothesized that functional polymorphisms in NAD(P)H quinone oxidoreductase 1 (NQO1) and superoxide dismutase 2 (SOD2), modulators of the effects of reactive oxidative species, can influence an individ...

No association between GSTM1 and GSTT1 genetic polymorphisms and susceptibility to opium sap dependence

Glutathione S-transferases (GSTs; EC: 2.5.1.18) are a ubiquitous family of eukaryotic and prokaryotic phase II metabolic isozymes. Genes encoding GSTM1 (OMIM: 138350), and GSTT1 (OMIM: 600436) are members of class mu and theta, respectively. The most common polymorphism in the GSTM1 is a deletion of the whole GSTM1 gene with a lack of enzyme activity. A homozygous deletion in the GSTT1 has also...

NAD(P)H:quinone oxidoreductase 1 (NQO1) Pro187Ser polymorphism and the risk of lung, bladder, and colorectal cancers: a meta-analysis.

UNLABELLED NAD(P)H:quinone oxidoreductase 1 (NQO1) is a cytosolic enzyme that catalyzes the two-electron reduction of quinoid compounds into hydroquinones, their less toxic form. A sequence variant at position 609 (C --> T) in the NQO1 gene encodes an enzyme with reduced quinone reductase activity in vitro and thus was hypothesized to affect cancer susceptibility. We conducted meta-analyses foc...

The relationship between NQO1 C609T and CAT C-262T genetic polymorphisms and the risk of age-related cataracts

Cataract is multi-factorial eye disease identified by the disturbance of the transparent ocular lens. There is significant evidence suggesting oxidative damage as a major cause of initiation and progression of numerous diseases including cataracts. NAD(P)H:quinone oxidoreductase 1 (NQO1; OMIM: 125860) and catalase (CAT, OMIM: 115500) are antioxidant enzymes that prevent cells from oxidative str...
